March 30, 1981 John Hinckley attempts to assassinate Ronald Reagan. Leaving a speaking engagement at the Hilton Hotel in Washington, DC, Reagan and three others are shot and wounded by John Hinckley Jr. NBC correspondent Judy Woodruff said that at least one shot was fired from a window of a room of the hotel, above Reagan's limousine. She later elaborated, saying a Secret Service agent had fired that shot. Hinckley had six bullets in his gun yet seven bullets were fired at Reagan. Official narrative says a ricochet off the presidential limousine hit Reagan. But it would appear that the seventh bullet was shot from a higher position. FBI investigations into the shooting have never been released. The FBI determined special explosive bullets, called Devastators, were fired. Doctors who removed the bullet from the President's lung did not know during the operation, that the bullet could have exploded at any second. Hinckley had no bullets in his possession when he was arrested, and none were found in his belongings at the Park Central hotel where he stayed. The shadow government did not care for Ronald Reagan. | Here we engineered full,
partially deleted (-29 nt), and fully deleted ORF8 into a SARS-CoV infectious
cDNA clone, strain Frankfurt-1. Replication of the resulting virus was compared in primate cell cultures as well as Rhinolophus bat cells made permissive for SARS-CoV replication by lentiviral transduction of the human angiotensin-converting enzyme 2 receptor. Cells from cotton rat, goat, and sheep provided control scenarios that represent host systems in which SARS-CoV is neither endemic nor epidemic. Independent of the cell system, the truncation of ORF8 (29 nt deletion) decreased replication up to 23-fold. The effect was independent of the type I interferon response. |
Leaderless genome attenuation of gene expression
| The establishment of persistent viral
infection is often associated with the
selection of one or more
mutant virus. For example, it has been found that an intraleader open reading frame (ORF) in genomic and subgenomic mRNA (sgmRNA) molecules is selected during bovine coronavirus (BCoV) persistence which leads to translation attenuation of the downstream ORF. Here, we report the unexpected identification of leaderless genomes, in addition to leader-containing genomes, in a cell culture persistently infected with BCoV. The discovery was made by using a head-to-tail ligation method that examines genomic 5'-terminal sequences at different times postinfection. Functional analyses of the leaderless genomic RNA in a BCoV defective interfering (DI) RNA revealed that (1) the leaderless genome was able to serve as a template for the synthesis of negative-strand genome, although it cannot perform replicative positive-strand genomic RNA synthesis, and (2) the leaderless genome retained its function in translation and transcription, although the efficiency of these processes was impaired. Therefore, this previously unidentified leaderless genome is associated with the attenuation of genome expression. Whether the leaderless genome contributes to the establishment of persistent infection remains to be determined. |
